DescriptionL-Leucine, also known as (S)-leucine or L-leucin, belongs to the class of organic compounds known as leucine and derivatives. Leucine and derivatives are compounds containing leucine or a derivative thereof resulting from reaction of leucine at the amino group or the carboxy group, or from the replacement of any hydrogen of glycine by a heteroatom. L-Leucine exists as a solid, possibly soluble (in water), and a very strong basic compound (based on its pKa) molecule. L-Leucine exists in all living species, ranging from bacteria to humans. L-Leucine can be converted into L-leucine through its interaction with the enzyme large neutral amino acids transporter small subunit 1. In cattle, L-leucine is involved in the metabolic pathway called the leucine stimulation ON insulin signaling pathway. L-Leucine is a potentially toxic compound.

Transcriptional activator. Binds the cAMP response element (CRE) (consensus: 5'-GTGACGT[AC][AG]-3'), a sequence present in many viral and cellular promoters. Cooperates with FOXO1 in osteoblasts to regulate glucose homeostasis through suppression of beta-cell production and decrease in insulin production. Regulates the induction of DDIT3/CHOP and asparagine synthetase (ASNS) in response to endoplasmic reticulum (ER) stress. In concert with DDIT3/CHOP, activates the transcription of TRIB3 and promotes ER stress-induced neuronal apoptosis by regulating the transcriptional induction of BBC3/PUMA.
