Phenotypes
Cataract, recessive, NID1-related
As reported by Murgiano et al. (2014), the candidate region (mentioned in the Mapping section above) contains 42 annotated and several uncharacterised genes, none of which are obvious functional candidate genes for this disorder. Analysis of whole-genome sequencing data from of one of the affected calves (~x13.5) and 44 other cattle from 15 breeds narrowed the field of positional candidate causal mutations down to one SNV in an uncharacterised...
Convulsions and ataxia, CACNA1A-related — Bovine familial convulsions and ataxia; cerebellar abiotropy
Reith et al. (2024): "Whole-genome sequencing was performed on the [Angus] sire, six affected and seven unaffectedpaternal half-sibling calves and combined with data from 135 unrelated controls. The sire and five of the six affected calves were heterozygous for a nonsense variant (Chr7 g.12367906C>T, c.5073C>T, p.Arg1681*) in CACNA1A. The other affected calves (N = 8) were heterozygous for the variant but it was absent in the other unaff...
Hypohidrotic ectodermal dysplasia, X-linked, EDA-related — Congenital hypotrichosis and anodontia defect; Ectodermal dysplasia; X-linked hypohidro...
Because of the obvious homology of this disorder with the homologous human disorder, Drögemüller et al. (2001) proposed that the bovine disorder be called by the name of its human homologue, which is now done in this catalogue. The earlier names are listed here as species-specific names [Frank Nicholas 20 June 2002].
Hypotrichosis, streaked — Streaked hairlessness
Murgiano et al. (2015) investigated a family of Pezzata Rossa cattle with 4 affected cows. The pedigree was suggestive of X-linked semi-dominant inheritance with lethality in hemizygous mutant males. Two autosomal segments on chromosome 7 and 14 also showed positive LOD scores in a linkage analysis and could not definitively be excluded. Whole genome sequencing of one affected cow at 28x coverage and comparison to the 1000 bull genomes' sequen...
Ichthyosis, ABCA12-related
In the words of Charlier et al. (2008): "a missense mutation in exon 39 (A5804G) resulting in an H1935R substitution in the fourth extracellular loop". The His (normal) form of the peptide is conserved in all vertebrates sequenced to date. (FN 080330) Whole-genome sequencing of the affected Shorthorn calf described by O'Rourke et al. (2017), and subsequent checking for deleterious variants in functional candidate genes, enabled Woolley et al. ...
Syndactyly — Mule foot disease; mulefoot; Haplotype HHM
Syndactyly has been reported in many breeds of cattle in many countries. Most of the documentation, however, concerns its occurrence in US Holsteins, where, as a result of the siring of more than 60,000 calves by a bull who was subsequently shown to be a carrier, the disorder attracted considerable attention (Anon., 1967). The possibility that artificial selection favouring heterozygotes may have contributed to the unacceptably high frequency ...
